Crhr2−/− male mice have impaired blood glucose clearance in response to insulin challenge. In ITT, blood glucose levels were measured by tail-vein sampling before (baseline; 0 min) and at 15, 30, 45, and 60 min after ip insulin administration in chow- and HFD-fed mice. Repeated-measure ANOVA followed by Sidak’s post hoc test was used to analyze ITT data. ac Male WT mice showed normal response to ITT challenge. HFD-fed Crhr2−/− male mice had higher blood glucose levels at all times examined and a 2-fold higher AUC. df Female WT and Crhr2−/− mice retained normal ITT responses on chow and HFD. n = 8/group/sex. g CRF2 receptor levels were ~ 1.5-fold higher in livers of WT male on HFD vs. chow. Expression levels were normalized to β-actin, a housekeeping gene. Student’s t test, n = 8/group
