Gating strategy for the determination of monocytes in the blood. First, living cells were selected based on size in the forward side scatter plot. Next all CD11b⁺ cells were selected, and within this population, all Ly6G⁻ cells were gated to excluded granulocytes. Within the remaining monocytes, classical, intermediate, and non-classical monocytes were distinguished using ly6C. Within these three subsets MHC2⁺ and CD80⁺ cells were selected.
